Beskrivelse: This antibody recognizes a glycoprotein of 110 kDa, which is identified as CD68. It is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. It reacts with myeloid precursors and peripheral blood granulocytes. It also reacts with plasmacytoid T cells, which are supposed to be of monocyte/macrophage origin. It shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Lymphomas are negative or show few granules.

Beskrivelse: CD2 interacts through its amino-terminal domain with the extracellular domain of CD58 (also designated CD2 ligand) to mediate cell adhesion. CD2/CD58 binding can enhance antigen-specific T cell activation. CD2 is a transmembrane glycoprotein that is expressed on peripheral blood T lymphocytes, NK cells and thymocytes. CD58 is a heavily glycosylated protein with a broad tissue distribution in hematopoietic and other cells, including endothelium. Interaction between CD2 and its counter receptor LFA3 (CD58) on opposing cells optimizes immune system recognition, thereby facilitating communication between helper T lymphocytes and antigen-presenting cells, as well as between cytolytic effectors and target cells.

CD1 proteins have been demonstrated to restrict T cell response to non-peptide lipid and glycolipid antigens and play a role in non-classical antigen presentation. CD1a is a non-polymorphic MHC Class 1 related cell surface glycoprotein, expressed in association with Beta-2 microglobulin. Anti-CD1a labels Langerhans cell histiocytosis (Histiocytosis X), extranodal histiocytic sarcoma, a subset of T-lymphoblastic lymphoma/leukemia, and interdigitating dendritic cell sarcoma of the lymph node. When combined with antibodies against TTF-1 and CD5, anti-CD1a is useful in distinguishing between pulmonary and thymic neoplasms since CD1a is consistently expressed in thymic lymphocytes in both typical and atypical thymomas, but only focally in 1/6 of thymic carcinomas and not in lymphocytes in pulmonary neoplasms. Anti-CD1a is reported to be a new marker for perivascular epithelial cell tumor (PEComa).

Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Carcinoids, neuroendocrine carcinomas of the skin, and oat cell carcinomas of the lung also express neurofilament.

Beskrivelse: Recognizes a protein of 55 kDa, identified as CD25 (Workshop IV; Code A27). CD25 is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R beta chain, p75) and CD132 (common gammachain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. This MAb recognizes the epitope B, which is located at residue 3-104 of CD25 and can effectively block IL-2 binding to CD25.

Beskrivelse: A disulphide-linked heterodimer, consisting of mb-1 (or CD79a) and B29 (or CD79b) polypeptides, is non-covalently associated with membrane-bound immunoglobulins on B cells. This complex of mb-1 and B29 polypeptides and immunoglobulin constitute the B cell Ag receptor. CD79a first appears at pre B cell stage, early in maturation, and persists until the plasma cell stage where it is found as an intracellular component. CD79a is found in the majority of acute leukemias of precursor B cell type, in B cell lines, B cell lymphomas, and in some myelomas. It is not present in myeloid or T cell lines. Anti-CD79a is generally used to complement anti-CD20 especially for mature B-cell lymphomas after treatment with Rituximab (anti-CD20). This antibody will stain many of the same lymphomas as anti-CD20, but also is more likely to stain B-lymphoblastic lymphoma/leukemia than is anti-CD20. Anti-CD79a also stains more cases of plasma cell myeloma and occasionally some types of endothelial cells as well.

Beskrivelse: Recognizes proteins of 33, 35 and 41 kDa, which are identified as various isoforms of CD74. Its epitope is localized in the extracellular domain of CD74. CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The CD74 molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74 is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, and dendritic cells and by various types of carcinomas. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.

Beskrivelse: This antibody reacts with human CD59, a 20 kDa glycosyl phosphatidyl-inositol (GPI)-anchored cell surface protein. CD59 regulates complement-mediated cell lysis, and it is involved in lymphocyte signal transduction. This protein is a potent inhibitor of the complement membrane attack complex, whereby it binds complement C8 and/or C9 during the assembly of this complex, thereby inhibiting the incorporation of multiple copies of C9 into the complex, which is necessary for osmolytic pore formation. CD59 is widely distributed on cells in all tissues. It inhibits formation of MAC, thus protecting cells from complement-mediated lysis. The expression of CD59 on erythrocytes is important for their survival. Genetic defects in GPI-anchor attachment, that cause a reduction or loss of CD59 and CD55 on erythrocytes produce the symptoms of the disease paroxysmal hemoglobinuria (PNH). It is useful for study on GPI-anchored proteins, PNH and CD59 functions.

CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.
