Beskrivelse: FLUOR DE LYS® Developer is an essential reagent for signal generation in HDAC or sirtuin assays done with the FLUOR DE LYS® substrate. An appropriate FLUOR DE LYS® family substrate, comprising an acetylated lysine side chain, is first incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes it so that, in a second step, treatment with the FLUOR DE LYS® Developer produces a fluorophore. Typically, an ‘inhibitor stop’ will be included in the 1x Developer dilution (e.g. Trichostatin A) for class I and II HDACs, nicotinamide for sirtuins).

Immobilised Papain offers the advantage of generating Fab and fragments without the need to remove the papain enzyme after digestion. Following papain digestion the Fab fragments are separated from undigested IgG and the Fc region with the supplied Protein A Spin Column. Protein A Resin binds the IgG and Fc molecules and the Fab are rapidly collected.

Beskrivelse: The F(ab)₂ Fragmentation kits are designed for the generation and isolation of F(ab)₂ fragments from IgG molecules. The kit utilises Immobilised Pepsin resin. Pepsin is a proteolytic enzyme that is routinely used for the generation of F(ab)₂ fragments from immunoglobulin G (IgG). One or more of the disulfide bonds that join the heavy chains in the hinge region are preserved, so the two Fab regions of the antibody remain joined together, yielding a divalent molecule (containing two antibody binding sites), hence the designation F(ab)₂. The light chains remain intact and attached to the heavy chain, whereas the Fc fragment is digested into small peptides. The Immobilised Pepsin offers the distinct advantage of eliminating enzyme contamination of the F(ab)₂ fragments.
